Changes of Platelet Count and Aggregation Fuction in Stored Blood at 4¡É


Abstract
This study was designed to investigate the sequential changes of platelet count and
platelet aggregation function in stored blood at 4¡É.
Blood obtained from healthy 5 persons was stored at the ACD-anticoagulated screw
capped tube. Platelet count was measured by Coulter counter S-Plus ¥± and platelet
function was evaluated by the aggregation function with ADP, epinephrine, ristocetin,
and collagen, respectively at 0 hours, 3, 6, 12, 24 hours and 1st, 2nd, 3rd, 5th, 7th, l0th
and 14th days after storage.
The results were obtained as follows.
1. Platelet counts were reduced to 73.3% at 3 day, 68.2% at 5th day and 39.3% at
14th day after storage.
2. Little change was shown in the aggregation pattern of ADP and ristocetin until 3
days after storage. Aggregation function was sustained about 50% at 5th day and
markedly reduced thereafter.
3. Aggregation function to epinephrine was immediately decreased after storage but
was sustained about 50% in 3rd or 5th day of storage.
4. Marked inhibition of aggregation function to collagen was shown. So, prolonged lag
phase and reduced aggregation function was observed in proportion to storage period.
